Glycerol Phenylbutyrate is a taste-masked, liquid prodrug of phenylbutyrate developed to improve the long-term treatment experience for patients with urea cycle disorders. Salt forms of phenylbutyrate can carry a strong taste and a high sodium load; glycerol phenylbutyrate addresses both by delivering three phenylbutyrate units on a glycerol backbone as a nearly odourless, tasteless oil.
After administration it is gradually hydrolysed to release phenylbutyrate, providing sustained nitrogen-scavenging activity and steadier ammonia control — which supports better adherence, especially in paediatric and long-term patients.
